Efficacy of Recombinant Human Erythropoietin




Abstract
Effect of recombinant human erythropoietin(rHu-EPO) on erythropoiesis were investigated in the normal and cisplatin- and acute hemorrhage-induced anemic rats and the hypoxia-induced and transfusion induced polycythemic mice. Normal rats were treated intravenously with rHu-EPO far 6 days axid the changes in the values of the red blood cells(RBC), hemoglobin(Hb), hematoarit(Hct), and reticulocytes were examined. Normal rats showed significant increases of all the hematolpgical values at doses rHu-EPO 250 and 1,250IU/§¸. In acute hemorrhagic-induced anemic rata, rHu-EPO(20 or 100IU/§¸) increased significantly erythrocytes, Hb, Hct, RBC and reticulocytes in dose dependent manners. Cisplatin-induced anemic rats showed significant increases of RBC, Hb, Hct and reticulocytes in dose dependent manners after rHu-EPO 200 or 400IU/§¸ treatment. In histopathological findings of kidney, cisplatin-alone treated rate revealed severe damages to proadmal convoluted tubule. However, the lesions in rate treated with rHu-EPO after cisplatin treatment were milder than those of ciaplatin- alone treated rata. The cisplatin-induced renal in juries and anemia were restored by rHu-EPO treatment. Mice were placed in hyposic chamber maintained at 12.6% O©ü for 7 days with free access to food and water for hypoxic exposure. The rHu-EPO(100IU/§¸) pre-and post-treatment in hypoxia-induced erythremic mice increased the value of RBC, Hb, Hct and reticulocytes. In transfusion-induced polycythemic mice, the values of RBC, Hb, Hct and reticulocytes were increased after a single dose of rHu-EPO. These results suggest that rHu-EPO might be useful for the therapy of anemia induced by acute blood loss or ciaplatin, and that rHu-EPO could induce erythropoiesis in polycythemia induced by hypoxia or transfusion.
